The College of Science for Women / University of Baghdad examined the Dissertation entitled (The role of several microRNA types in mucin 1,4,16 expressions in infertile Iraqi females under IVF program) of the student (Israa Mahmood Majeed Ahmed), in Partial Fulfillment of the Requirements for the Degree of Doctor   in Biology/ Zoology.

 

Study the role of several micro RNA types in gene expression of mucin 1,4,16 genes in infertile Iraqi females under IVF program in order .

This study is designed to determine the role of several micro RNA types in gene expression of mucin 1, 4, and 16 genes in infertile Iraqi females under an in vitro fertilization (IVF) program in order to improve the IVF program. The study included (128) females under the IVF program divided into two significant parts first, 84 infertile females: – 26 successful implantations, 58 failure failed implantation under an in vitro fertilization program, and second 44 fertile females under IVF as control divided into two minor subgroups, 21 successful implantations, and 23 failed implantation. Quantitative Real-Time PCR (q-PCR) was performed to measure the gene expression of miR-146a-3p, miR-146a-5p, miR-485-5p, miR-210-HG,miR-1226, and mucin-1,4, and16 genes

 

The study concluded that is up regulation of miR-146a-5p, miR-210 HG, miR-485-5p directly regulate mucin-1 gene expression. Mucin1 has a high negative linkage with miR-146a-5p and miR-485-5p, and a strong positive correlation with microR-210. In infertile females undergoing IVF, implantation failure is associated with a substantial elevation in mucin-1 expression. The infertility cases under the IVF program with fertile failure implantation cases related to significant down-regulation of mucin-4, except successful fertile cases recorded significant up-regulation. The up-regulation of miR-210 HG, miR-485-5p may contribute to an increased chance of failed implantation in females undergoing IVF therapy that targets mucin-4 expression. Mucin4 has a significant positive association with miR-210 and miR-485-5p in infertile females and a negative correlation with miR-485-5p in fertile females. Elevated mucin-16 levels significantly influence successful implantation in females participating in the IVF program, up regulation of miR-1226 and miR-210-HG are potential inhibitors of embryo implantation failure in infertile females, up regulation of miR-146a-3p regulates unsuccessful embryo implantation, and down-regulation of miR146a-3p regulate successful implantation in fertile females by targeting mucin-16 gene expression because a strong negative correlation between mucin-16 with miR-1226 and positive correlation with miR-210 in infertile females and a significant negative correlation between mucin-16 with miR-146a-3p and miR-485-5p in fertile females.
