The study was supervised by <strong>Asst. Prof. Dr. Shatha Thanoon Ahmed<\/strong><\/p>\n<\/div><\/div><\/div><div class=\"relative\"><div id=\"cz_97039\" class=\"cz_97039 cz_line tal\"><\/div><\/div><div class=\"clr\"><\/div><div class=\"cz_gap clr \" style=\"height: 50px\"><\/div><div id=\"cz_81239\" data-arrows='{\"open\":\"fa fa-angle-down\",\"close\":\"fa fa-angle-up\"}' class=\"cz_81239 cz_acc clr\"><div><div id=\"cz_76098\"><span class=\"cz_acc_child\"><i class=\"cz-acc-i cz-acc-icon fa czico-052-three-2\"><\/i><div>Objectives of the thesis<\/div><\/span><div class=\"cz_acc_child_content clr\">[vc_column_text css=&#8221;&#8221;]The thesis investigated the antibacterial and antibiofilm effects of <em>Lactobacillus plantarum<\/em>\u00a0crude extract (<em>L.p<\/em>Ex) and its chitosan nanoparticle formulation (<em>L.p<\/em>ExCsNPs) against clinical isolates of <em>P. aeruginosa<\/em>\u00a0and <em>S. aureus<\/em>. A total of 122 clinical specimens were collected from patients in Baghdad hospitals, resulting in 36 isolates of <em>P. aeruginosa<\/em>\u00a0(29.5%) and 25 isolates of <em>S. aureus<\/em>\u00a0(20.4%). PCR confirmed the identity of isolates using housekeeping genes (rpsL and 16S rRNA). Biofilm production was observed in all isolates with varying degrees of strength. Antibiotic susceptibility tests revealed high resistance in both bacterial species, most notably to Penicillin (100%) in <em>S. aureus<\/em>\u00a0and Meropenem (63.8%) in <em>P. aeruginosa<\/em>.[\/vc_column_text]<\/div><\/div><div id=\"cz_78368\"><span class=\"cz_acc_child\"><i class=\"cz-acc-i cz-acc-icon fa czico-052-three-2\"><\/i><div>Content of the thesis<\/div><\/span><div class=\"cz_acc_child_content clr\">[vc_column_text css=&#8221;&#8221;]<em>L.p<\/em>Ex was encapsulated into chitosan nanoparticles using the ionic gelation method. Characterization using UV-Vis, FTIR, SEM, EDX, and XRD confirmed the production of spherical, crystalline nanoparticles with smooth surfaces and a diameter range of 41.21\u201363.18 nm. MIC results showed that L.pExCsNPs had a significantly lower MIC (196.15 \u00b5g\/ml) compared to L.pEx (15625 \u00b5g\/ml), indicating enhanced antibacterial efficacy. Anti-biofilm testing at sub-MIC concentrations demonstrated superior inhibition by L.pExCsNPs, reducing biofilm formation by 60% in <em>P. aeruginosa<\/em>\u00a0and 38% in <em>S. aureus<\/em>, compared to 41% and 20% respectively with the crude extract.[\/vc_column_text]<\/div><\/div><div id=\"cz_23796\"><span class=\"cz_acc_child\"><i class=\"cz-acc-i cz-acc-icon fa czico-052-three-2\"><\/i><div>thesis recommendations<\/div><\/span><div class=\"cz_acc_child_content clr\">[vc_column_text css=&#8221;&#8221;]Gene expression of the biofilm-associated genes algD (<em>P. aeruginosa<\/em>) and icaA (<em>S. aureus<\/em>) was assessed using qRT-PCR. L.pEx treatment resulted in mean fold changes of 0.0633 (algD) and 0.7320 (icaA), while L.pExCsNPs treatment showed stronger inhibition with fold changes of 0.0015 (algD) and 0.1830 (icaA).
